I am using a Fishman Tripleplay MIDI guitar interface with AmpliTube, but when I open the program the only option for input is 'bulit-in mic', which gives awful feedback. There is no drop-down option for Tripleplay or anything else (I even tried my Oxygen 25 keyboard controller). Please advise. Thank you.

Both Triple play and the oxygen 25 are midi controllers and only send midi notes and controls.
Amplitube needs an audio signal form your guitar, and you need a digital audio interface attached to your computer (PC or Mac). This interface should appear as an ASIO (PC) or Core audio audio input in Amplitube
The built-in mic is not a proper HI-Z audio input for your guitar

Yes AmpliTube is a (64-bit) standalone program and plug-in (Audio Units, VST 2, VST 3, AAX.)
