How do i use the Irig pro io to play through phone to AmpliTube and then out to a guitar amp?

You would need to send the headphone output on iRig Pro I/O (assuming you don't mean iRig Pro Duo I/O) to your guitar amp. This requires a cable, like the Mono Output Adapter in IK's iLine kit and available separately:
